Intended Use:
The kit is used for in vitro quantitative detection of human leukocyte antigen B27 gene (including B2702, B2704, and B2705 subtypes) in human whole blood samples.
Ankylosing spondylitis (AS) is a rheumatic disease characterized by the involvement of the central axis and peripheral joints, and chronic inflammation and the formation of new bones. The disease is more common in adolescents. Severe joint dysfunction can occur within 10 years of onset. When the spine, sacroiliac joints and hip joints develop lesions, the lesions are irreversible. Among them, human leukocyte antigen-B27 (HLA-B27) is considered to have a strong correlation with AS, that is, the positive rate of HLA-B27 in AS patients is as high as 90%. The kit developed by our company can detect the B2702, B2704, and B2705 subtypes related to AS, but it cannot distinguish between different genotypes. The test results of this kit are for the auxiliary diagnosis of AS. The test results shall not be used as the only indicator of the patient's condition evaluation. The patient's clinical manifestations and other testing methods must be combined to make a comprehensive analysis of the condition.
Principle of Test:
A pair of HLA-B27 specific primers are selected and combined with specific probes. The probes can specifically bind to a DNA template in the middle of the primer amplification region. In the process of PCR extension, the exonuclease activity of Taq enzyme will cut off the 5'end fluorescent groups from the probes, freeing them in the reaction system, thus breaking away from the shielding of the 3'end fluorescence quenching groups, which can receive light stimulation and emit fluorescence that can be detected by the instrument, this kit uses FAM channel, so as to achieve the automatic detection of HLA-B27 nucleic acid in closed reaction system.

The probes can specifically bind to a DNA template in the middle of the primer amplification region. In the process of PCR extension, the exonuclease activity of Taq enzyme will cut off the 5'end fluorescent groups from the probes, freeing them in the reaction system, thus breaking away from the shielding of the 3'end fluorescence quenching groups, which can receive light stimulation and emit fluorescence that can be detected by the instrument, The internal control adopts VIC channel, so as to realize the detection process monitoring in closed reaction system, which can effectively monitor the occurrence of false negatives.
Storage Condition & Validity:
All reagents should be stored at -20ºC±5 ºC. The validity period of the reagents is 12 months.

Sample Requirements :
1. Sample: human whole blood samples.
2. Collection: 3~5mL of venous blood is extracted from the subject with a disposable sterile syringe, and injected into a glass tube containing EDTA
anticoagulant. The glass tube is immediately reversed gently and mixed for 5~10 times, so that the anticoagulant and venous blood are fully mixed.
3. Storage: samples can be stored at 2~8ºC for no more than 24 hours; under -20ºC for no more than 3 months; under -70ºC for long-time, but
repeated freeze-thaw should be avoided.
4. Transportation: It is transported in a foam box with ice to seal for transportation.
