Intended Use:
The kit is used for in vitro quantitative detection of EBV nucleic acid in human whole blood samples.
EBV belongs to the genus of herpesvirus gamma subfamily Lymphocytovirus, also known as human herpesvirus type 4. Diseases caused by EBV include: infectious mononucleosis, African childhood lymphoma (Burkitt lymphoma) and nasopharyngeal carcinoma.
The laboratory tests of EBV include:
1) Virus isolation and culture;
2) Serological testing;
3) Real-time fluorescence quantitative polymerase chain reaction, etc.
The kit developed by our company is suitable for the auxiliary diagnosis of EBV infection. The test results of this kit are for clinical reference only and cannot be used as the basis for diagnosis or exclusion of cases.
Principle of Test :
The highly conserved sequences of EBV are selected to design specific primers and Taqman probes. The probes can specifically bind to a DNA template in the middle of the primer amplification region. In the process of PCR extension, the exonuclease activity of Taq enzyme will cut off the 5'end fluorescent groups from the probes, freeing them in the reaction system, thus breaking away from the shielding of the 3'end fluorescence quenching groups, which can receive light stimulation and emit fluorescence that can be detected by the instrument, so as to achieve the automatic detection of EBV nucleic acid in closed reaction system.
A pair of conserved gene-specific primers are selected as the internal standard in the kit and combined with specific probes. The probes can specifically bind to a DNA template in the middle of the primer amplification region. In the process of PCR extension, the exonuclease activity of Taq enzyme will cut off the 5'end fluorescent groups from the probes, freeing them in the reaction system, thus breaking away from the shielding of the 3'end fluorescence quenching groups, which can receive light stimulation and emit fluorescence that can be detected by the instrument, the internal control adopts VIC channel, so as to realize the detection process monitoring in closed reaction system, which can effectively monitor the occurrence of false negatives.
Storage Condition & Validity:
All reagents should be stored at -20ºC±5 ºC. The validity period of the reagents is 12 months.
The unused reagents can be sequentially cryopreservation without affecting their stability. The regents could be preserved for 30 days at -20ºC±5 ºC after repeated thawing and freezing three times. The opened reagents should be placed at room temperature under the condition of no more than 8 hours. The products should be transport by ice box or refrigerated truck. Simulated transport tests indicate that the stability and validity could not be influenced by transport, but the transportation time should not exceed 7 days. Applicable Instrument:
Gentier 96E real time PCR system from Xi'an Tianlong Science and Technology Co. Ltd., Roche Light Cycler 480 real-time fluorescence quantitative PCR instrument or similar apparatus .
Sample Requirements:
1. Sample: whole blood samples.
2. Collection: 3~5mL of venous blood is extracted from the subject with a disposable sterile syringe, and injected into a glass tube containing EDTA anticoagulant. The glass tube is immediately reversed gently and mixed for 5~10 times, so that the anticoagulant and venous blood are fully mixed.
3. Storage: samples can be tested immediately, stored at 2~8ºC for no more than 48 hours; -20ºC±5 ºC for no more than 3 months; under -70ºC for long-time, but repeated freeze-thaw should be avoided.
4. Transportation: use a curling pot with ice or a foam box with ice to seal for transportation.
